Mouse anti-Human CDw328 antibody, clone 5-386 recognizes human CDw328, a type I transmembrane glycoprotein and member of the Siglec (sialic acid-binding Ig-like lectin) family, designated Siglec-7.
It was originally identified as an inhibitory NK cell receptor (NKR) and negative regulator of NK activation, attributed to ITIM recruitment of SHP-1 phosphatase.
Target Information
CD328 (Siglec7)는 세포 간 시알산 의존적 결합을 매개하는 접착 분자로 추정됩니다.
- α2,3- 및 α2,6-결합된 시알산에 선호적으로 결합
- 이중 시알산당지질에도 결합 (예: disialogalactosyl globoside 등)
- 시알산 인식 부위는 동일 세포 표면의 시알산과의 상호작용에 의해 가려질 수 있음
면역 반응에서, 리간드 유도성 티로신 인산화 후 SH2 도메인을 가진 인산가수분해효소(SHP-1 등)를 모집하여 신호 전달을 억제합니다.
이로 인해 NK 세포의 세포독성 활성이 억제되며, 조혈 과정(hemopoiesis)에도 관여할 수 있습니다.
또한 CD34+ 세포 전구체의 골수계(myelomonocytic) 분화 및 백혈병성 골수 세포 증식을 억제하는 것으로 알려져 있습니다 (in vitro).
