Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). CD11b is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It serves as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, playing key roles in cell-cell and cell-matrix interactions.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells; use at 5 µL (0.8 µg) per test.

Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent structures minimizing spillover and spread impacts.

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia. CD11b/CD18 acts as a receptor for ICAMs, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating critical adhesion functions. Expression varies by tissue and cell type, increasing during monocyte maturation. Dysregulation is linked to systemic lupus erythematosus.
