Product Specific Information

The ICRF44 monoclonal antibody recognizes human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). It is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. CD11b acts as a receptor for intercellular adhesion molecules (CD54, CD102, CD50) and iC3b, playing key roles in cell-cell and cell-matrix interactions.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended usage: 5 µL (0.8 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL buffer per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with CD18 (integrin beta-2) to create Mac-1 (CR3). It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
Expression levels vary by cell type and tissue, increasing with monocyte maturation. CD11b is commonly used as a microglial marker in nervous tissue. Abnormal CD11b function is linked to diseases such as systemic lupus erythematosus.
