Product Specific Information

The HA58 monoclonal antibody reacts with human CD54 (ICAM-1), a 90–110 kDa transmembrane glycoprotein expressed on monocytes, lymphocytes, and endothelial cells. CD54 expression increases upon lymphocyte activation, and its interaction with CD11a plays a key role in inflammatory responses.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Flow cytometry of normal human peripheral blood cells

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample (regardless of number of NovaFluors) to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring engineered fluorescent structures that minimize spillover and spread.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein with five immunoglobulin-like extracellular domains, a transmembrane region, and a cytoplasmic tail. It binds integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1), and serves as a receptor for rhinovirus.

Expression:
Detected on activated endothelial, epithelial, fibroblast, chondrocyte, B and T lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils. Expression increases during inflammation and is found in some carcinoma and melanoma cells. Soluble ICAM-1 is present in plasma and elevated in inflammatory conditions.
