Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin receptor family. It plays a central role in signal transduction, T cell differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC class I molecules.
The CD8α chain is critical for MHC-I binding and is also expressed as disulfide-linked homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
Engagement of MHC-I/peptide complexes by CD8 triggers recruitment of Lck kinase, promoting lymphokine production, motility, and activation of cytotoxic T lymphocytes (CTLs), which are key in pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
