Target Information
Janus kinase 3 (JAK3) phosphorylates members of the STAT (signal transduction and activators of transcription) family.
JAK3 is predominantly expressed in immune cells and mediates signaling in response to activation of interleukin receptors.
Upon stimulation by cytokines or related factors (e.g., interferon, interleukins), JAKs are activated via phosphorylation at two adjacent tyrosine residues.
Activated JAKs phosphorylate STAT proteins, which dimerize and translocate to the nucleus to regulate transcription of interferon-stimulated genes.
JAK2 is essential for IFN-γ receptor complex initiation, while JAK1 acts as an amplifier. Studies suggest JAK2’s role may be complemented by Tyk2 and JAK3 depending on their positioning within the receptor complex.
