Target Information
PIAS4 functions as an E3-type small ubiquitin-like modifier (SUMO) ligase, stabilizing the interaction between UBE2I and its substrates, and acts as a SUMO-tethering factor. It plays a key role as a transcriptional coregulator in multiple signaling pathways, including STAT, p53/TP53, Wnt, and steroid hormone pathways. It is involved in gene silencing and mediates sumoylation of CEBPA, PARK7, HERC2, MYB, TCF4, and RNF168. In Wnt signaling, it represses LEF1 and enhances TCF4 transcriptional activities by promoting their sumoylation. It also enhances MTA1 sumoylation and may participate in paralog-selective sumoylation. [UniProt]
