• Specificity: MHM24 binds to the CD11a subunit of lymphocyte function-associated antigen. It binds to both the high- and low-affinity forms of the antigen.
• Preparation: Spleen cells from BALB/c mice immunised with EBV-transformed human lymphoblastoid cells were fused with P3-NSUlAg4.1 myeloma cells and the resulting hybridomas were screened for antibody production by a radioimmunoassay on T and B cell lines.

Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the alphaLbeta2 integrin. CD11a is implicated in activation of LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAMs 1–3. It also functions in lymphocyte costimulatory signaling.
CD11a is expressed on leukocyte plasma membranes in a low-affinity conformation, which changes to high-affinity upon stimulation. This transition supports tight binding to ICAMs, facilitating immune cell interactions, migration, and differentiation.
Blocking CD11a function has therapeutic implications in inflammatory diseases. Humanized anti-CD11a antibody Efalizumab (Raptiva) and simvastatin binding to CD11a are examples of immunomodulatory approaches.
Two transcript variants encoding different isoforms of CD11a have been found. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
