Target Information
This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant).
These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two of four conserved cysteine residues. Binding of chemokines to CXCR3 induces leukocyte traffic responses such as integrin activation, cytoskeletal changes, and chemotactic migration.
Inhibition by Bordetella pertussis toxin suggests coupling with Gi-class G proteins. The receptor undergoes phosphorylation-dependent internalization upon activation, followed by recycling after dephosphorylation. CXCR3 is highly expressed in effector/memory T cells and inflamed tissues, mediating recruitment of inflammatory cells.
Thus, CXCR3 is a potential therapeutic target for inflammatory disease modulation. Multiple transcript variants encoding different isoforms have been identified.
