Target Information
KLRB1 (Killer cell lectin like receptor subfamily B, member 1, CD161) protein is classified as a type II membrane protein with an external C terminus. It is expressed on nearly all NK cells and a subset of CD3+ T cells. KLRB1 forms a homodimeric cell surface protein (40–44 kDa) and acts as an inhibitory receptor on NK cells.
Activation of KLRB1 stimulates acid sphingomyelnase/SMPD1 and increases intracellular ceramide, as well as activation of AKT1/PKB and RPS6KA1/RSK1 kinases. It also promotes T cell proliferation via anti-CD3 stimulation. KLRB1 functions as a lectin binding to Gal-α(1,3)Gal and N-acetyllactosamine epitopes, and interacts with CLEC2D/LLT1 to inhibit NK cell-mediated cytotoxicity and interferon-gamma secretion.
