단백질의 유비퀴틴화(ubiquitination)는 비정상적이거나 수명이 짧은 단백질을 분해 대상으로 지정하는 중요한 세포 내 메커니즘입니다.
이 과정에는 E1(ubiquitin-activating enzymes), E2(ubiquitin-conjugating enzymes), E3(ubiquitin-protein ligases) 세 가지 효소군이 관여합니다.
UBE3B 유전자는 E3 ubiquitin-conjugating enzyme 계열에 속하며, E2 효소로부터 유비퀴틴을 받아 표적 단백질로 전달합니다.
이 단백질의 긴 isoform의 C-말단에는 HECT(homology to E6-AP C-terminus) 도메인이 존재하며, 유비퀴틴 전달의 촉매 부위로 작용합니다.
짧은 isoform은 HECT 도메인이 결여되어 있어 E2 효소와 결합하지 못할 가능성이 있습니다.
이 유전자는 서로 다른 isoform을 암호화하는 대체 스플라이싱(transcript variants)을 통해 다양한 형태로 발현됩니다.
