Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, receptor-associated kinase에 의해 인산화되어 핵으로 이동 후 전사 인자로 작용합니다.
STAT3의 활성화에는 CREB-binding protein 등의 공동활성화 인자가 필요하며, Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화됩니다.
Tyrosine 705의 JAK1 및 JAK2에 의한 인산화는 STAT3의 이합체 형성, 핵 이동 및 DNA 결합 활성에 필수적입니다.
STAT3 유전자는 인간 염색체 17q에 위치하며, 세포 성장, 분화, 아폽토시스 등 다양한 세포 과정에 관여합니다.
Rac1은 STAT3의 활성을 조절하며, PIAS3는 STAT3의 특이적 억제 단백질로 알려져 있습니다.
세 가지 alternative splicing 변이체가 보고되었으며, STAT3 유전자 결손 생쥐는 초기 배아 단계에서 치명적입니다.
