Target Information
Activins are dimeric growth and differentiation factors belonging to the transforming growth factor-beta (TGF-beta) superfamily.
They signal through a heteromeric complex of receptor serine kinases, including type I (I and IB) and type II (II and IIB) receptors.
These receptors are transmembrane proteins composed of:
- Ligand-binding extracellular domain with cysteine-rich region
- Transmembrane domain
- Cytoplasmic domain with serine/threonine specificity
Type I receptors are essential for signaling, while type II receptors are required for ligand binding and type I receptor expression.
Upon ligand binding, type I and II receptors form a stable complex, leading to phosphorylation of type I receptors by type II receptors.
Mutations in Activin A are associated with fibrodysplasia ossificans progressiva.
