Background
DNA methylation is a major epigenetic modification in eukaryotic genomes, essential for mammalian development. Human proteins MECP2, MBD1, MBD2, MBD3, and MBD4 form a family of nuclear proteins characterized by a methyl-CpG binding domain (MBD). Except for MBD3, these proteins specifically bind to methylated DNA. MECP2, MBD1, and MBD2 can repress transcription from methylated promoters. The MBD2 protein may mediate biological consequences of methylation and is also reported to function as a demethylase, activating transcription by reversing methylation-induced gene silencing. Two transcript variants encoding different isoforms have been identified for this gene (RefSeq, Feb 2011).
