Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
Recommended usage: 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

Blocking Buffer Usage:
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
Add 5 µL per sample to antibody cocktails prior to combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). Absent on non-hematopoietic cells and tissues, CD45 regulates lymphocyte development and antigen signaling via Src-family kinases. Multiple isoforms arise from alternative splicing, differing in extracellular domains. CD45RA expression varies among lymphoid subtypes and physiological states.

CD45 is essential for T- and B-cell receptor signaling, modulates cytokine receptor signaling via JAK kinases, and influences cell survival through integrin-mediated pathways and apoptosis regulation.
