Product Specific Information

The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a TNFR family member expressed on mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
It regulates B cell maturation and Ig isotype switching, protects B cells from apoptosis, and promotes proliferation. Interaction with CD154 (gp39) is crucial for T–B cell crosstalk and immune regulation.
The 1C10 antibody exhibits agonistic activity in vitro and in vivo.

Target Information

CD40 antigen is a single-chain glycoprotein belonging to the TNFR/NGF superfamily, homologous to CD30.
Expressed on all B cells except plasma cells, and on certain epithelial cells, carcinomas, and dendritic cells.
CD40 is essential for immune and inflammatory responses, including:

• T cell-dependent immunoglobulin class switching
• Memory B cell development
• Germinal center formation

AKNA transcription factor regulates CD40 expression for cell interactions. TNFR2 acts as a signal transduction mediator.
CD40–CD154 interaction is linked to amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease.
Two alternatively spliced transcript variants exist.
