Target Information

The antimicrobial protein BPI is a 55 kDa protein found in the primary (azurophilic) granules of human neutrophils and also on the surface of neutrophils, small intestinal, and oral epithelial cells.
BPI is a bactericidal compound present in polymorphonuclear cells (PMN) and eosinophils. It binds strongly to the lipid A region of lipopolysaccharides (LPS) in gram-negative bacteria, leading to multiple anti-infective activities such as:

  • Cytotoxicity through damage to bacterial membranes
  • Neutralization of bacterial LPS
  • Opsonization to enhance phagocytosis by neutrophils

BPI may play a key role in LPS-triggered airway disease, as airway epithelial cells express and produce BPI. Inflammation induced by LPS can contribute to rapid airflow decline, a severe complication of hematopoietic cell transplantation.
Additionally, a 21 kDa recombinant fragment, rBPI21, has shown protective effects against invasive pneumococcal disease by binding to pneumolysin, a gram-positive bacterial toxin.
