Target Information
PIF1 is a single-stranded (ss) DNA-dependent ATPase and DNA helicase that unwinds DNA in a 5′ to 3′ direction. It plays a critical role in DNA replication and telomere length maintenance in Saccharomyces cerevisiae. The PIF1 gene is highly conserved from yeast to humans. Mutations in PIF1 lead to telomere elongation, indicating its function as a catalytic inhibitor of telomere formation and elongation. Human PIF1 binds telomeric DNA and inhibits telomerase activity, counteracting the RecQ homolog Sgs1 helicase activity. Degradation of PIF1 is mediated by the ubiquitin–26S proteasome pathway.
