Target Information

Myelin Protein Zero (MPZ), also known as Zero, is a Type I integral membrane glycoprotein that mediates adhesion of spiraling wraps of the myelin sheath, ensuring stable synaptic transmission. MPZ constitutes approximately 50% of total protein in the peripheral myelin sheath, contributing to its structural integrity. It facilitates compact myelin wrapping via glycine zipper packing interface-dependent dimer and tetramer formation. Mutations, such as G134R, disrupt multimer formation and are linked to demyelinating neuropathies, including Charcot-Marie-Tooth disease. The cytoplasmic domain undergoes serine and tyrosine phosphorylation, particularly during peak nerve myelination. MPZ transcript levels are moderate in brain, abundant in thymus, and highest in white matter of the CNS.
