Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and has also been identified as a co-receptor for HIV-2 via envelope glycoprotein gp120, facilitating viral fusion. CXCR4 binds to the chemokine CXCL12/SDF-1, triggering intracellular calcium increase and MAPK pathway activation. Additionally, CXCR4 acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling. The receptor plays key roles in hematopoiesis, cardiac development, vascularization of the gastrointestinal tract, and neuronal survival in the CNS. It also mediates inflammatory responses through LPS binding and TNF secretion. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
