Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and has also been shown to mediate HIV-2 binding and fusion. CXCR4 serves as the receptor for the C-X-C chemokine CXCL12/SDF-1, triggering intracellular calcium increase and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP levels.
CXCR4 is involved in hematopoiesis, cardiac ventricular septum formation, and gastrointestinal vascularization by regulating endothelial cell branching and remodeling. In the CNS, it contributes to hippocampal neuron survival and cerebellar development. CXCR4 can bind bacterial LPS, mediating inflammatory responses including TNF secretion by monocytes. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection of human target cells.
