Target Information
p27 Kip1 functions as a negative regulator of G1 phase progression and is proposed to mediate TGFβ-induced G1 arrest. It is a key regulator of cell cycle progression and a potent inhibitor of cyclin E- and cyclin A-CDK2 complexes. p27 Kip1 interacts with cyclin D-CDK4 complexes, influencing their assembly, stability, and activation. Depending on its phosphorylation state or stoichiometry, p27 Kip1 can act as either an inhibitor or activator of cyclin D-CDK4 complexes. The protein has an approximate molecular weight of 27 kDa, and its degradation — triggered by CDK-dependent phosphorylation and subsequent ubiquitination by SCF complexes — is essential for the transition from quiescence to the proliferative state.
