Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was initially identified as a surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates cell proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in Hodgkin’s disease and other CD30+ lymphomas, acting as a costimulatory molecule in thymic negative selection. It is expressed not only in Hodgkin’s and Reed-Sternberg cells but also in certain non-Hodgkin’s lymphomas, virus-infected T and B cells, and activated normal T and B cells.

In T cells, CD30 is expressed on subsets that produce Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. The soluble form (sCD30) reflects Th2 immune response. CD30 interacts with TRAF2 and TRAF5 to activate NF-kappaB signaling, functioning as a positive regulator of apoptosis and limiting autoreactive CD8 effector T cell proliferation.

Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
