The Human SPARC (Osteonectin) Simplex ProcartaPlex Kit measures SPARC (osteonectin) protein and is designed to be combinable with other Simplex kits, allowing users to create custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (Cat. No. EPX010-10420-901) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of a protein to quantitate multiple targets simultaneously on a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and deliver analyzed results in about four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 μL sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads are internally dyed with precise proportions of red and infrared fluorophores, creating 100 spectrally unique bead signatures identifiable by Luminex xMAP systems (Luminex 200, FLEXMAP 3D, and MAGPIX). Each bead is conjugated with antibodies specific for a single protein target, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a magnetic bead rather than adsorbed to a microplate well, allowing reagents to remain in solution. The Luminex instrument uses two lasers — one to identify bead signatures and another to quantify RPE fluorescence proportional to protein concentration. This enables profiling of more targets using less sample in the same time as a standard ELISA.

ProcartaPlex multiplex panels are available across six species (human, mouse, rat, nonhuman primate, porcine, and canine).
