Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human Adiponectin. Anti-Human Adiponectin-specific antibody was purified by affinity chromatography employing an immobilized Human Adiponectin matrix.

Sandwich ELISA:
To detect Human Adiponectin by sandwich ELISA (using 100 µL/well), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human Adiponectin (500-P239BT) as a detection antibody, allows the detection of at least 2000–4000 pg/mL of Recombinant Human Adiponectin.

Western Blot:
To detect Human Adiponectin by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human Adiponectin is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

Adiponectin is a protein hormone secreted exclusively by adipocytes that regulates the metabolism of lipids and glucose. It plays a role in various physiological processes including homeostasis and obesity. Decreased levels of adiponectin are associated with obesity, insulin resistance, and hyperinsulinemia (type 2 diabetes). In mice, adiponectin has been shown to improve insulin signaling, ameliorating insulin resistance. Through activation of AMP kinase and PPAR-alpha, adiponectin increases fatty acid transport, oxidation, and dissipation in skeletal muscle, reducing intramyocellular lipid levels and improving insulin signaling.
