Target Information

Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in various diseases such as cancer. Members of the Bcl-2 family are critical regulators of apoptosis, characterized by conserved Bcl-2 homology (BH) domains.
Bcl-rambo is a novel, widely expressed member localized to mitochondria in mammalian cells. Its overexpression induces apoptosis, which can be blocked by co-expression of inhibitor of apoptosis proteins (IAPs) such as XIAP, cIAP1, and cIAP2.
Bcl-rambo shows homology to anti-apoptotic members but contains a unique 250 amino acid insertion preceding its C-terminal membrane anchor. This region can induce apoptosis more efficiently than the BH domains, suggesting Bcl-rambo’s role in additional pro-apoptotic pathways.
