Description:
The eBioGHI/61 monoclonal antibody reacts with human CD163 (Haemoglobin Scavenger Receptor, M130), a macrophage scavenger receptor family member. CD163 is expressed by tissue macrophages, circulating blood monocytes, and may be found on hematopoietic progenitor cells. It functions as a high-affinity scavenger receptor for the hemoglobin-haptoglobin complex, clearing hemoglobin from the blood.

CD163 binding triggers IL-6 and IL-10 production. Cytokines such as IL-6 and IL-10 upregulate CD163 expression, while proinflammatory stimuli (e.g., LPS, TNFα) induce shedding of soluble CD163.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.5 µg) per test (final volume 100 µL). Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, a member of the scavenger receptor cysteine-rich superfamily, and a receptor for the hemoglobin-haptoglobin complex. It protects tissues from oxidative damage and may mediate iron recycling via endocytosis and heme breakdown.

CD163 is expressed on human monocytes and macrophages, especially in late inflammation phases. It is present on all CD14⁺ monocytes, most CD64⁺ monocytes, and shows higher expression on CD16⁺ monocytes. Expression is upregulated by IL-10, IL-6, and dexamethasone, and downregulated by LPS or PMA through shedding into plasma or supernatant.

Binding of hemoglobin/haptoglobin complexes is calcium- and pH-dependent, showing higher affinity for multimeric haptoglobin (HP1F phenotype). CD163 signaling involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and IL-6/CSF1 secretion.
