Product Specific Information
Description:
The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C), which are expressed on all nucleated cells and associated with β2-microglobulin. It recognizes a non-polymorphic epitope shared among HLA-A, B, and C products and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkey.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported:
- Flow cytometric analysis
Applications Tested:
- Pre-titrated and tested for flow cytometry using normal human peripheral blood cells
- Recommended use: 5 µL (0.6 µg) per test (in 100 µL final volume)
- Cell number: 10⁵–10⁸ cells/test
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample prior to combining antibodies with cells.
- For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
Technology:
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for controlled fluorescence signatures and minimal spectral spillover.
