Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, it dissociates from accessory proteins, translocates to the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

The AR signaling pathway is essential for the development and function of male reproductive organs such as the prostate and epididymis, and also influences nonreproductive organs including muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase activity. Abnormalities in AR signaling are associated with prostate cancer, Kennedy’s disease, and male infertility. Mutations in this gene can cause complete androgen insensitivity (CAIS).
