Target Information
Alpha 2-Macroglobulin (a2M) is an approximately 720-kDa homotetrameric glycoprotein composed of four identical ~180 kDa subunits. It shares with other alpha-macroglobulins (e.g., complement components C3, C4, and pregnancy zone protein PZP) a remarkable ability to bind a wide range of ligands, functioning as a humoral defense barrier against foreign peptides in plasma. Alpha 2M captures virtually any proteinase, acting as a panprotease inhibitor. In Alzheimer’s disease (AD) brains, Alpha 2M is localized to diffuse amyloid plaques, suggesting an important role in AD pathogenesis.
