Target Information
Myosin phosphatase target subunit 1 (MYPT1), also known as the myosin-binding subunit of myosin phosphatase, is a regulatory component controlling actin-myosin interactions downstream of Rho GTPase signaling. RhoA activates Rho-kinase, which phosphorylates and inactivates MYPT1, thereby increasing myosin light chain (MLC) phosphorylation and promoting smooth muscle contraction and actin-myosin interaction in nonmuscle cells. Overexpression of RhoA or its active form enhances phosphorylation of MYPT1 and MLC. Multiple transcript variants encoding different isoforms have been identified.
