Target Information
Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity (encoded by the TERT gene) and an RNA component that serves as a template for the telomere repeat.
Telomerase expression is normally repressed in postnatal somatic cells, leading to progressive telomere shortening and cellular senescence. Deregulation of telomerase expression may contribute to oncogenesis. Studies in mouse suggest telomerase also participates in chromosomal repair, as de novo synthesis of telomere repeats may occur at double-stranded breaks.
Alternative splicing of the TERT gene produces multiple isoforms, some of which are incompletely characterized, and this splicing is thought to regulate telomerase activity.
