Target Information

Crystallins are divided into two classes: taxon-specific (enzyme) and ubiquitous. The ubiquitous class forms the major proteins of the vertebrate eye lens, maintaining its transparency and refractive index. Lens fiber cells lose their nuclei during development, so crystallins are synthesized once and retained throughout life, making them extremely stable.

Mammalian crystallins are categorized into alpha, beta, and gamma families, with beta and gamma forming a superfamily. Alpha and beta are further divided into acidic and basic groups. Crystallins contain four homologous motifs, a connecting peptide, and N- and C-terminal extensions.

Gamma-crystallins are monomeric, highly symmetrical proteins lacking connecting peptides and terminal extensions, and are differentially regulated after early development. The encoded protein of this gene, initially considered a beta-crystallin, is monomeric and more similar to gamma-crystallins. It is the most significant gamma-crystallin in adult eye lens tissue. Mutations or aging-related changes in gamma-crystallins are associated with cataract formation.
