Target Information
The human UNG gene encodes both nuclear (UNG2) and mitochondrial (UNG1) forms of uracil-DNA glycosylase. These forms arise through alternative splicing and different promoter usage (PA and PB).
UNG1 and UNG2 cDNAs are similar in size (2061 and 2058 bp), and their mRNAs are not resolved as distinct species in gel electrophoresis.
UNG2 differs from UNG1 by 44 amino acids at the N-terminal sequence, which is not required for catalytic activity.
The primary role of UNG proteins is to repair mutagenic U:G mispairs caused by cytosine deamination—UNG2 removes misincorporated dUMP residues.
Expression of UNG1 and UNG2 varies by cell and tissue type and is cell cycle regulated, being higher in proliferating cells.
UNG1 is a 304 amino acid protein, and UNG2 is a 313 amino acid protein.
