Target Information
Peroxisome proliferators are non-genotoxic carcinogens that exert their effects through interaction with nuclear hormone receptors known as peroxisome proliferator-activated receptors (PPARs). These ligand-dependent intracellular proteins stimulate transcription of specific genes by binding to DNA sequences upon activation. PPARs are activated by compounds such as clofibric acid, nafenopin, and WY-14,643, as well as certain fatty acids. They induce transcription of acyl coenzyme A oxidase and cytochrome P450 (CYP450) A6 via specific response elements. PPARs form heterodimers with retinoid X receptor (RXR) alpha, similar to other nuclear hormone receptors.
