Target Information
DR5 (Apo2, TRAIL-R2, TRICK2, KILLER)는 TRAIL에 의해 유도되는 세포사멸(apoptosis)을 매개하는 death domain을 포함한 수용체입니다. TNF-receptor superfamily의 구성원으로, 세포 내 death domain을 포함하고 있으며, TNFSF10/TRAIL/APO-2L에 의해 활성화되어 세포사멸 신호를 전달합니다. FADD 결핍 생쥐 연구에서 FADD 단백질이 DR5 매개 세포사멸에 필수적임이 확인되었습니다. DR5는 두 가지 이형체(transcript variant)와 하나의 비암호화 전사체를 가지며, 과발현 시 apoptosis를 유도하고 NF-kappa-B를 활성화합니다. 림프구와 비장에서 높은 수준으로 발현되며, DR5 기능 이상은 편평세포암종 및 확산성 림프구증후군 등과 관련이 있습니다.
