Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and is a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking cell cycle transitions (G1→S, G2→M) after DNA damage to allow repair.
In the cytoplasm, p21 exerts anti-apoptotic effects by inhibiting caspase 3 and the apoptotic kinases ASK1 and JNK.
This dual role makes p21 both a tumor suppressor and an oncogene depending on context.
