Target Information
Beta-catenin is an adherens junction (AJ) protein originally identified as a component of cell-cell adhesion structures. AJs are essential for the creation and maintenance of epithelial cell layers by regulating cell growth and adhesion between cells. Beta-catenin interacts with the cytoplasmic domain of E-cadherin, linking it to alpha-catenin, which anchors the E-cadherin complex to the actin cytoskeleton.
Beta-catenin also associates with cytoskeletal complexes containing adenomatous polyposis coli protein and microtubules, participating in signaling pathways involving tyrosine kinases, phosphatases, and Wnt/Wingless. This interplay regulates morphogenesis and cell proliferation.
It is expressed in hair follicle cell types, basal and peripheral matrix cells, and outer/inner root sheath cells. Pathological roles have been identified in pilomatrixoma, medulloblastoma, colorectal, ovarian, and other solid tumors (breast, colon, liver, lung, gastric, prostate, skin). In the nucleus, beta-catenin co-activates Lef/Tcf transcription factors, stimulating genes like cyclin D and c-myc that drive proliferation and tumorigenesis.
