Target Information
Activator of S phase kinase (ASK, DBF4) is a regulatory subunit of the human Cdc7 kinase complex, playing a critical role in the G1/S cell cycle transition. Increased expression of ASK in late G1 contributes to elevated Cdc7 kinase activity at the G1/S boundary. ASK interacts with replication origins, suggesting Cdc7 may trigger S phase by activating replication initiation complexes. Human ASK levels fluctuate during the cell cycle, paralleling Cdc7 activity. The Cdc7-ASK kinase complex regulates DNA replication initiation by targeting MCM2 protein. Nuclear localization and chromatin binding of Cdc7 and ASK are cell cycle–dependent, increasing at late G1 phase. A functional link may exist between ASK and the tumor suppressor menin (mutated in MEN1), implicating ASK in cell proliferation control.
