Target Information
This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-g-inducible 10 kDa protein), Mig (monokine induced by interferon-g), and I-TAC (interferon-inducible T cell chemoattractant).
These chemokines belong to the CXC subfamily, characterized by a single amino acid between the first two conserved cysteines.
Binding of these chemokines induces leukocyte trafficking, integrin activation, cytoskeletal changes, and chemotactic migration.
Inhibition by Bordetella pertussis toxin indicates coupling with Gi-class G proteins.
Signal transduction mechanisms are similar to other chemokine receptors, involving phosphorylation-dependent receptor internalization and recycling.
CXCR3 is highly expressed in effector/memory T cells and in T cells within inflamed tissues.
IP10, Mig, and I-TAC production by local cells suggests a role in recruiting inflammatory cells.
This receptor is a potential therapeutic target for inflammatory diseases.
Multiple transcript variants encoding different isoforms have been identified.
