This antibody reacts with CD3 epsilon and is predicted to react with birds and all mammals based on sequence homology.
Heat-mediated antigen retrieval using Tris/EDTA (pH 9) is recommended for staining of paraffin sections.
A suggested positive control for immunohistochemical applications is tonsil tissue.
Rat anti-Human CD3, clone CD3-12, raised against a peptide representing an invariant cytoplasmic sequence within the CD3 epsilon chain, recognizes human CD3 epsilon.
Target Information
The CD3 subunit complex is crucial in transducing antigen-recognition signals into T-cell cytoplasm and regulating TCR complex expression.
T-cell activation involves CD3 gamma, delta, epsilon, and zeta subunits, which are structurally related members of the immunoglobulin superfamily encoded on human chromosome 11.
CD3 subunits associate with cytoplasmic signal transduction molecules via a double tyrosine-based motif.
CD3 antigens are expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.
Decreased T lymphocyte percentages may occur in some autoimmune diseases.
Defects in the CD3 gene are associated with CD3 immunodeficiency.
