Target Information

COX2 converts arachidonate to prostaglandin H2 (PGH2), a committed step in prostanoid synthesis, including inflammatory prostaglandins.
This conversion involves two reactions:

1. Cyclooxygenase (COX): arachidonate → prostaglandin G2 (PGG2)
2. Peroxidase: PGG2 → prostaglandin H2 (PGH2)

COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, and is up-regulated in pathological conditions (e.g., cancer, inflammation).
Up-regulation is linked to increased cell adhesion, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 facilitates prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance.
COX2 is naturally inhibited by calcitriol (active Vitamin D) and repressed by glucocorticoids.
It is also the target of NSAIDs such as aspirin, which reduce pain and inflammation.
