Product Specific Information

A recommended positive control tissue for this antibody is Seminoma, though other positive tissues may also be used.
This antibody is intended for laboratory professional use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue using manual qualitative IHC testing.

It recognizes a muco-protein of 38–43 kDa, identified as Podoplanin (PDPN). Podoplanin localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells. As a regulator of lymphatic endothelium, it likely contributes to maintaining the unique morphology of podocytes.

Podoplanin is selectively expressed in lymphatic endothelium, lymphoangiomas, Kaposi sarcomas, and certain angiosarcomas with lymphatic differentiation. It is also a sensitive and relatively specific marker for epithelioid mesothelioma, useful for distinguishing mesotheliomas or mesothelial cells from pulmonary carcinomas.

Staining and Control Recommendations

  • Use with formalin-fixed, paraffin-embedded sections.
  • Pretreatment with heat-induced epitope retrieval or enzymatic retrieval is recommended.
  • IHC staining visualizes antigens through sequential antibody and enzyme reactions, followed by chromogenic substrate detection.
  • Results should be interpreted microscopically to assist in differential diagnosis.

A positive tissue control must be included with each staining procedure. Positive control tissues should contain both positive and negative staining regions and be processed identically to patient samples.
Controls with weak positive staining are preferred to detect subtle changes in antibody sensitivity or reagent degradation.

Negative controls (internal or external) should also be used to confirm the absence of non-specific staining. Any specific staining in negative controls invalidates the results.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type transmembrane glycoprotein (40–43 kDa) expressed in various normal and tumor cells, especially lymphatic endothelial and follicular dendritic cells.

Podoplanin acts as a ligand for CLEC-2 and is associated with tumor invasiveness and metastasis. It is involved in cell migration, metastatic spread, and tumor invasion. Expression can be induced via Src activation of Cas and has been observed on Th17 cells and a subset of CD4+ PBMCs.

Dysfunction of Podoplanin is linked to conditions such as lymphangioma and subglottic neoplasm, and it may serve as a marker for lung injury due to its physiological function.
