This antibody is supplied in a ready-to-use formulation. Recommended positive control tissue includes Seminoma, though other tissues may also be suitable.
It is designed for professional laboratory use in detecting Podoplanin in formalin-fixed, paraffin-embedded tissues using manual qualitative IHC methods.

This antibody recognizes a muco-protein of 38–43 kDa, identified as Podoplanin (PDPN). It localizes to stromal cells of peripheral lymphoid tissue and thymic epithelial cells. As a regulator of lymphatic endothelium, Podoplanin may play a role in maintaining podocyte morphology. It is selectively expressed in lymphatic endothelium, lymphoangiomas, Kaposi sarcomas, and angiosarcomas with lymphatic differentiation.
Podoplanin is also a sensitive and specific marker for epithelioid mesothelioma, useful in panels distinguishing mesotheliomas from pulmonary carcinomas.

Sections should be formalin-fixed and paraffin-embedded. Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, with intermediate washing steps. The chromogen reaction produces visible staining interpreted under a light microscope.

A positive tissue control must accompany each staining procedure. Controls should contain both positive and negative elements to verify staining accuracy.
External positive controls should be freshly prepared and processed similarly to patient samples. Controls with weak positive staining are ideal for detecting subtle reagent degradation or instability.
Negative controls (internal or external) verify specificity and background staining; if positive staining occurs in negative controls, patient results are invalid.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type glycoprotein (~40–43 kDa) expressed in many tumors and normal cells, particularly lymphatic epithelial cells and follicular dendritic cells.
It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelium. Podoplanin acts as a ligand for CLEC-2, correlating with tumor invasiveness and metastasis.
It is involved in cell migration, metastasis formation, and tumor invasion. Expression is induced via Src activation of Cas, and recent evidence indicates presence on Th17 cells.
Dysfunction of Podoplanin is linked to lymphangioma and subglottic neoplasm, and it has been proposed as a marker for lung injury.
