Target Information
Survivin (IAP4)은 Inhibitors of Apoptosis Protein (IAP) 유전자 패밀리 중 가장 작은 구성원으로, 세포 신호전달, 세포분열, 대사 등 다양한 세포 기능에 관여합니다. 대부분의 정상 조직에서는 발현이 낮으나, 거의 모든 인간 암 조직에서 높은 발현을 보입니다. Survivin은 단일 BIR 도메인을 가지며, C-말단 RING finger 도메인이 없습니다.
이 단백질은 G2/M 세포주기 단계에서 발현이 높으며, 유사분열 시 방추체의 미세소관과 결합하여 세포사멸 억제 기능을 수행합니다. Survivin-미세소관 상호작용이 붕괴되면 caspase-3 활성 증가와 함께 세포사멸이 유도됩니다.
또한, 뇌 손상 후 성체 랫의 신경세포(astrocyte, neuron)에서 높은 발현을 보이며, NeuN 및 PCNA와의 공발현이 관찰됩니다. 이는 신경세포의 증식 반응 조절에 관여할 가능성을 시사합니다.
4가지 전사 변이체가 확인되었으며, 이 중 3가지의 full-length 서열이 알려져 있습니다. 암세포에서 survivin의 과발현은 세포사멸 억제 및 비정상적인 세포분열을 유도할 수 있습니다.
