Product Specific Information
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Crosslinking via immobilized UCHT1 triggers intracellular signaling leading to activation and proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (defined as staining a sample in 100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.
Alexa Fluor 660 emits at 690 nm and is suitable for spectral cytometers, multiplexed with APC, Alexa Fluor 647, and Alexa Fluor 700.

Target Information
The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by genes on chromosome 11. These subunits contain cytoplasmic tails involved in signal transduction via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. CD3 expression abnormalities are linked to certain autoimmune diseases and immunodeficiencies.
