Target Information

The human AF9 (MLLT3) gene is a common fusion partner with the ALL1 (MLL) gene at 11q23, resulting in the translocation t(9;11)(p22;q23). The AF9 gene spans over 100 kb and includes two breakpoint cluster regions (BCRs):

  • BCR1 within intron 4 (site A)
  • BCR2 spanning introns 7 and 8 (site B)

AF9 contains structural elements such as a DNA topoisomerase II cleavage site and DNase I hypersensitive site within intron 7 (BCR2). Two scaffold-associated regions (SAR1 in intron 4 and SAR2 encompassing exons 5–7) flank these cleavage sites. These regions share structural features with the MLL breakpoint cluster region, supporting a model of DNA breakage and repair–mediated recombination between MLL and AF9.
