Target Information
In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is a critical component of transcriptional regulation, primarily influenced by the acetylation of nucleosomal histones. Acetylation of lysine residues in histone tails induces allosteric changes that increase DNA accessibility to transcription factors. Conversely, deacetylation is associated with transcriptional silencing.
Mammalian HDAC8, isolated from human kidney, is a histone deacetylase homologous to other HDACs but with distinct tissue distribution. HDAC8 localizes to the nucleus and plays roles in tissue development and cancer etiology.
