Target Information

Interferon gamma (IFN-γ, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and natural killer cells in response to antigens and cytokines. It is a dimeric glycoprotein composed of two 146 amino acid subunits (~45 kDa functional homodimer). On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

Human IFN-γ exhibits species-specific activity and does not cross-react with mouse IFN-γ. Functions include:

  • Antiviral and tumor antiproliferative activity
  • Induction of MHC class I and II
  • Macrophage activation
  • Enhancement of immunoglobulin secretion by B lymphocytes

Activation occurs via binding to IFN-γ receptor I and II, triggering the JAK-STAT pathway. Human IFN-γ shares ~40% sequence homology with mouse IFN-γ. It is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN. Mutations in the IFN-γ gene are associated with aplastic anemia.
