Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2).
Foxp3 is a 49–55 kDa transcription factor essential for regulatory T cell (Treg) function.
Immunoblotting mapped the epitope to amino acids 75–125 of mouse Foxp3.
Optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00).
Use of other fixation and staining buffers is not recommended.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining and flow cytometric analysis of mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Recommended use ≤0.25 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Careful titration recommended for optimal performance.

Fluorophore Information:
PerCP-eFluor 710 emits at 710 nm, excited by 488 nm blue laser.
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Sensitive to photo-induced oxidation—protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis.
It is highly expressed in CD25⁺CD4⁺ regulatory T cells, low in CD4⁺CD25⁻ cells, and absent in CD4⁻CD8⁺ T cells.
FOXP3 defects cause scurfy phenotype in mice and IPEX syndrome in humans.
Transcript variants encoding different isoforms have been identified.
In human cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
